OP, you stated you swap coils and it stayed with cylinder #5?
Did you change/inspect the plugs?
My misfires with a tune have all been plug related. Could not make it misfire with stock firmware. Dropped in the Cobb and misfire city.
Changed the plugs and misfires are gone.
